A customer wants to know that this product is specific to SGLT1 (That is not reacted with SGLT2). Isthis product tested inbeing specific to SGLT1? If you have tested data, please send me. Or if not data, could you tell me somethingproving to be specific to SGLT1?
We would like to know whether SGLT1 Positive Control (PC-SGLT) are developed from insect sources or baculovirus expression systems.
I"m currently working on 5 different breast cancer cell lines and I"m using yours standard protocol for WB in that particular Antibodies, with 40ug of protein instead 60 (I have less nonspecific bands). My WB shows the band at approx. 58kDa in each cell line. I"ve checked UniProt database for SGLT1 and indeed there are 2 isoforms of this protein. The difference is about 120aas on the beginning of the protein sequence. I"m just curious why my antibodies recognizes only one isoform (the shorter protein) instead of two though the immunogen covers the opposite end of SGLT1 sequence (C-terminus) presents in both isoforms. For now, I"m pretty confused what protein I see in my WB.

1. Our SGLT1 antibodies, SGLT-101AP and SGLT-112AP, are epitope specific antibodies. They are raised against synthetic peptides that are unique to the SGLT1 protein and are not present in SGLT2. These antibodies do not react to random peptide in ELISA and in Dot Blot assays. These antibodies are affinity purified on immobilized antigen based affinity chromatography. Based on these observations, they will not cross react with SGLT2 proteins.
We would like to know whether SGLT1 Positive Control (PC-SGLT) are developed from insect sources or baculovirus expression systems.
1. SGLT1 Positive Control (PC-SGLT1.m and PC-SGLT.C) are not obtained from insect sells, SF9 cells or other cells. It is a partially purified protein obtained from normal enterocytes. The preparation is in SDS-Sample buffer and does not contain any preservatives. The shelf life is 1 year at -20⁰C and 6 months at 4⁰C.

1. We have two epitope specific antibodies to SGLT1. The antibody SGLT-101AP is a N-epitope (with respect to the other epitope) and SGLT-112AP is a C-epitope downstream from the SGLT-101AP. Both antibodies were tested on intestinal brush boarder membranes (BBM) and appears tolabel the high MW isoform of SGLT1. We did not see the smaller variant of SGLT1 in BBM. It is possible that smaller variant is abundant in breast cancer cells and high MW variant is not expressed in these cells. These observations yet to be confirmed using PC-SGLT1 samples or using recombinant SGLT1 proteins and compare them with renal BBM and intestinal BBM samples.
